No — both TIFF and PNG are lossless formats. The conversion is completely lossless at the same bit depth. Every pixel in the PNG output is identical to the corresponding pixel in the TIFF source. The only exception is if you choose to convert from 16-bit to 8-bit depth, which reduces precision by discarding the lower 8 bits of each channel — this is visible in very fine gradients as a slight banding artefact.
Use "Auto (match TIFF)" in most cases — the PNG will have the same bit depth as the TIFF. Use "8-bit" if you need the PNG for web use (browsers and most apps display 8-bit PNGs; 16-bit PNGs are supported by Chrome and Firefox but not all image viewers). Use "16-bit" when working with scientific data, HDR photography, or medical imaging where the full tonal precision of the TIFF must be preserved.
PNG uses lossless Deflate compression — the output file size depends on the image content. Photographic images with complex, irregular pixel patterns compress poorly in PNG (often only 20–40% size reduction from uncompressed). TIFF files that use LZW or ZIP compression may actually be similar in size to a well-compressed PNG. If file size is a priority, convert to JPEG (lossy) instead of PNG. For logos, diagrams, and flat-colour illustrations, PNG compresses very well.
